In this experiment, human PBMCs were analyzed for transcription factor expression across CD4 and CD8+ T cell subsets. A large multi-color panel was required, such that we wanted to include a third color for the red laser. Tonbo Bioscience's redFluor 710 was selected as an alternative to Alexafluor 700 and similar reagents. We stained with CD45RA-redFluor 710 to differentiate naive and memory lymphocytes. This antibody provided excellent resolution of CD45RA expression levels in CD8+ gated T cells, actually enabling us to see 3 different levels of expression high, intermediate, and low.

Results Summary

The CD45RA-redFluor 710 antibody fit well in a complex multi-color panel involving human lymphocytes and retained good resolution of 3 levels of CD45RA expression in CD8+ T cells. This antibody outperformed other antibodies that we have tried which only showed 2 distinct levels of expression. Notably, the fluorescence intensity and resolution was preserved with aldehyde fixation and subsequently permeablization.
