CD294 or commonly known as CRTH2 is a chemo-attractant receptor molecule expressed on Th2 cells but not on Th1 cells. It is seven trans-membrane G-protein coupled receptor which is involved in leukocyte migration. It is also present in a small CD8+ T cell population. The aim of this experiment was to evaluate the increased expression of CD294 in CD4+ T cells ( Th2 population) compared to CD8+ T cells.
Flow Cytometry
Human PBMC or peripheral blood
PBMC’s (stimulated vs. non-stimulated) were stained with 1ul (1:100) of anti-CD294 APC cy7 antibody (the same cells were simultaneously stained for a multi-color panel with appropriate antibody dilutions according to the experiment design). Cells were kept for 30 mins. at room temperature. After incubation cells were washed twice with 1XPBS and re-suspended in 500ul of staining buffer. Cells were then analyzed by flow cytometry where mononuclear cells were gated according to FSC and SSC and expression of CD294 on CD4 or CD8 T cells were measured.

Expression of CD294 was increased in CD4+ T cell (Th2) population compared to CD8+ cells in an autoimmune disease indication that CRTH2 is responsible for inducing leukocyte infiltration of the Th2 sub-population only.
