The Lens Culinaris agglutinin is one of the few lectins, which efficiently bind to the vascular endothelium of the chick embryo. Immunofluorescent staining of chick embryo vasculature is important in developmental biology research and in cancer related studies involving human tumor cells (or tumor cells of any other species since chick embryo is a naturally immunodeficient host). The Fluorescein Lens Culinaris Agglutinin is especially helpful to illuminate chick embryo vasculature when tumor cells are tagged with a "red" immunofluorescent protein, for example the Tomato protein. The image depicts tumor-associated vasculature in the embryo grafted with B16F10 murine melanoma cells.

Starting Material

Chick embryo incubated for 12-15 days.

Protocol Overview

Chick embryo was inoculated with Fluorescein LCA at 1:20 dilution in PBS.The chorioallantoic membrane (CAM) was excised and visualized in immunofluorescent microscope without fixation.

Tips

Use 100-200 ul of 1:20 dilution of LCA. Wait 5-10 min and proceed with the microscopy.

Results Summary

Fluorescein LCA produces excellent staining of chick embryo CAM vasculature.
