We wanted to determine whether IL-6 was secreted by dendritic cells isolated from 2 different mouse models after treatment with DMF. We chose this kit because it was cost effective and had been cited in many research papers.

Mouse cell culture supernatant
I first separated the cells from the supernatant by centrifugation. The supernatant was then serially diluted . We followed a standard sandwich ELISA protocol of which the kit contained all of the components. I added the capture antibody provided, followed by my sample and then the detection antibody. Streptavidin-HRP and Substrate solution followed for 20 mins each. All wells were wash 3x between solutions. The optical density of each well was determined using a microplate reader set to 450 nm.
Optical density should be measured immediately after stop solution is added to avoid high background
We found varying concentrations of IL-6 at different time points measured. Results were consistent when repeated.
