Our lab analyzes murine CD4+ T cells and their role in inflammatory processes. Isolation of CD4+ T cells is often accomplished using CD4+ magnetic bead isolation. The use of these beads requires a DETACHaBEAD reagent to remove the cells.
Cell isolation/positive selection of cells
Murine lymph nodes and spleen
Homogenize isolated mouse lymph nodes and spleen and perform positive selection using CD4+ magnetic beads. After incubating sample(s) with magnetic beads, resuspend bead/cell pellet in 1mL cell culture media. Add DETACHaBEAD at the equivalent volume of the magnetic beads that was used. Incubate cells at room temperature for 45 minutes. Spin samples down to pellet beads. Remove supernatant containing purified cells.
Adjust volume of beads and DETACHaBEADS from the recommended volume suggested by the supplier to maximize cell isolations
A very pure (~98%) CD4+ cell population is achieved when using this product following isolation. Cells are very viable and healthy. 1-2x10^7 cells are isolated from mouse lymph nodes and spleen.
