Our lab routinely performs IHC on mouse CNS tissue to assess axonal damage and neuroinflammation. To visualize staining, we use the Vector HRP kits and develop sections with DAB. This kit delivers quality results and is easy and simple to use.
Immunohistochemistry
Coronal mouse brain sections
1) Incubate sections in primary antibody then wash 2a) Incubate sections in anti-mouse IgG biotinylated secondary antibody solution2b) While sections incubate in secondary solution, mix A+B solution, incubate at room temperature for 1hr3) Wash sections following secondary antibody incubation then incubate sections in A+B solution for 30min at room temperature4) Wash sections then incubate in DAB5) Coverslip in Permount
Be sure to treat sections with hydrogen peroxide to prevent background staining from endogenous peroxidases
Staining will have a rusty brown and can be visualized via light microscopy
