In our lab we do many flow cytometry experiments with human cells for which we use panels of mouse anti-human antibodies. Since we stain the same sample with many different antibodies we need to set up compensation controls to use in the set up of the cytometer in order to be able to distinguish the differently stained populations of cells

mix a drop of positive and a drop of negative control beads in a FACS tube. Stain with single antibody (same staining protocol as used when staining cells). Set up compensation controls on the cytometer before collecting any cell samples
Can prepare the compensation controls in advance or use them for several experiments in the future.
Easily distinguished positive and negative control. Very helpful for FACS experiments where several colors are used in a single sample
