Our lab uses QIAGEN's PCR purification kit to 'clean up' our PCR products (i.e. eliminate excess primers, nucleotides, and enzymes) prior to confirming product size using agarose gel electrophoresis. Like all of QIAGEN's products that we use, the kit is time efficient, reliable, and easy to use -- all while providing us with great results.

Protocol Overview

Add 5x volume of Buffer PBI to each PCR sample, and check that each sample is yellow. Apply each sample to a column (included in the kit) and centrifuge for 1 minute. Discard the flow through and complete the wash/spin steps (i.e. Buffer PE). Elute each DNA sample into a clean tube with Buffer EB.

Tips

Use colorless tubes -- must interpret pH based on color of sample. If a higher concentration of DNA product is required, incubate for 1-5 minutes with buffer EB prior to spin/DNA elution.

Results Summary

PCR product accuracy and purity can be determined following gel electrophoresis (identifying product bands of the targeted size).

I recommend this product for individuals conducting cloning and/or sequencing experiments. The kit is incredibly easy to use and very reliable, as it provides great clean-up of DNA PCR products for downstream applications (like cloning, sequencing).
