Our lab studies HIV and immune aspects that can protect against infection. We have developed an in vitro system of generating latent CD4 HIV infected cells and are exploring reagents that will reverse latency. We need a very sensitive way to pick up virus reactivation, and in addition to RT-qPCR, we perform p24 ELISA to pick up virus that is being produced in the supernatant.

Protocol Overview

We follow the kits protocol religiously. We prepare biotin-acceptor beads + anti-HIV 24 antibody right before the assay, as well as the streptavidin donor beads. We do not dilute our samples, however we are measuring very low levels of infection. Only when infection is high, we run again with diluted samples.

Tips

Pay attention to what your levels on infection will be. Might need to run a test just to determine the dilution needed for samples
