Was using this antibody as part of a panel to look at localization of a protein within nuclear substructures. This was used in mouse, rat, and human cell lines. The mouse and rat cell lines displayed high background and diffuse non-specific staining, but the human cells showed distinct cajal bodies as puncta.

Results Summary

The antibody stained puncta assumed to be Cajal bodies in all cell lines tested. There was a large amount of diffuse (background?) staining present in the mouse and rat lines, while the human lines showed very bright and distinct puncta without any diffuse staining. The epitope is mapped to the human Coilin, so this may be the issue. Works great for human cells (H4 and HEK), but did not work as clearly for the mouse and rat lines (N2a and PC12).

This antibody is definitely worth a shot if you are looking to mark Cajal bodies in human cells/tissue. I wouldn't recommend this for staining non-human cells/tissues, as the high background is distracting.
