We are interested in enhancing the killing capacity of HIV-specific CD8 T cells by activating them via TCR specific MHC-peptide ligands, and co-stimulatory molecules. We use purified CD8 T cells from naive donors with with HLA types, such as B27 and A2, that have been described to mount potent anti-HIV responses to perform in vitro killing assays. The data shown here represents results from a positive control, where CD8 T cells were activated with PMA and ionomycin and then stained for degranulation markers, granzyme B, perforin, and different cytokine production.
