Our lab breeds various genetically modified mouse strains and we frequently need to genotype them before we use them. This kit allows us to quickly extra DNA from a small tissue snip so we can genotype them fast.

Application

PCR small tissue pieces for mouse genotyping

Starting Material

Make a mastermix for all samples before aliquotting.

Protocol Overview

A small piece of mouse tail or ear punch is added to a 0.2mL PCR tube. A master mix containing 25uL buffer, 0.5uL primer, 1uL Taq (all per sample) are added to a separate tube. Water is added to make a total of 50uL mix per sample. 50uL are then added to each PCR tube with the tail. The tube are vortexed to mix and added to a thermo-cycler and run on a program specific to the melting temps of the primers. After PCR run, 2uL of Proteinase K is added to the reaction to get rid of protein contaminants.

Tips

Make a mastermix with everything except tissue sample before aliquotting.

Results Summary

We are able to run a PCR on whole tissue sample without previous DNA extraction (using other kits, you must digest the tissue and extract the DNA before running the PCR). This allows us to quickly PCR a tissue sample and run on an electrophoresis gel for genotyping.
