We have various genetically modified mouse lines which require genotyping via PCR. After PCR we need to run the amplified product on an agarose gel before staining. This system makes the perfect size and thickness gel for that. It comes with a mold and combs for making the gel. The system itself does not slide around or spill when leaned on.

2% agarose gel

Protocol Overview

A 2% agarose gel (in 1x TAE) is poured into the gel mold and the correct size comb is added.PCR amplification is run with mouse tissue sample. The dried gel is removed from the mold and added to the electrophoresis system tray and the amplified DNA is added to each well. The system is connected to power and gel is run at 120V for 30 minutes in 1x TAE running buffer. The gel is then removed and stained for DNA detection.

Tips

Make sure you connect the (+) and (-) cords to the correct channel.

Results Summary

We are able to quickly and easily run amplified PCR products using a size gradient so we can identify target genes and genotype our animals smoothly/

Gel box is steady, will not slide around, tray has a fluorescent ruler.

Gel mold will crack if other things are place on top of it.
