NeuN is often used as a marker of neurons for histology experiments. Earlier antibodies against neun have been mouse monoclonal antibodies that are incompatible with experiments where other markers only have mouse based antibodies. This is the second neun antibody raised in rabbit that I've found [abn78 also from EMD Millipore being the first I've tried]. This antibody stains in the expected pattern and specificity with a good signal to noise ratio. It does seem to stain a lot of the soma as well as the nucleus, but this is not a problem for most experiments where it is being used as a control/reference as a general neuronal marker.

Rat brain and mouse brain - PFA fixed then frozen
1:500 dilution in blocking buffer for 48 hours at 4C
5% donkey serum 1% BSA in 1x PBS with 0.01% Triton X100
1:500 dilution in blocking buffer for 16 hours at 4C [JaxIR 488 conjugated donkey anti rabbit]
N/A
Fluorescence microscopy (488 channel)
Staining with this antibody is primarily localized to neurons. there is strong signal and minimal background. some staining of the soma and projections of the neruons occurs in addition to the nucleus.
None
It works
Possibly more soma staining than some other NeuN antibodies
This antibody is another useful option when a neuronal marker is needed and antibodies raised in mouse are not an useful.
