In the present study, Cdc6 proteins were detected in diabetic and non-diabetic rat kidneys maintained at different nutritional environment. In brief, rats of treatments were sacrificed at correct time point and kidney were harvested. Whole tissue extracts were prepared and protein was estimated. An equal amount of proteins were resolved in SDS-PAGE and western transferred in nitrocellulose membrane. Further, blots were probed with cdc6 antibody procured from Abcam. Selection of antibody from Abcam was based on information provided in antibody flyer.

Overnight at 4 0C (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:1000)
5 % Skimmed milk in TBS with 0.1% Tween 20 (for 1 hr at RT)
1 hr at RT (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:5000)

cdc6 antibody has given a highly intense band of cdc6 proteins at nearly 68 kDa. Cdc6 protein band was in duplicate and very sharp and clear.
