Aim of the present study to detect ASC protein levels in retinas of diabetic rats fed with or without phytoextract. Eyes from rats of all groups were enucleated, retinas were isolated and retinal extract were prepared. An equal amount of proteins were resolved in SDS-PAGE and immunoblotted for ASC protein using its specific ASC antibody (N-15) obtained from Santacruz Biotechnology. Selection of antibody was based on previous experience with same antibody.

Rat retinal tissue protein lysate
Overnight at 4 0C (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:500)
5 % Skimmed milk in TBS with 0.1% Tween 20 (for 2 hr at RT)
1 hr at RT (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:4000)

Poor quality ASC protein band with low signal were observed after chemiluminescent detection. ASC antibody bound with a lot of non specific proteins in rat samples.
ASC protein were detected
Lots of non specific bands were observed
Overall poor quality santacruz biotech antibody to detect ASC proteins in rat tissue samples.
