Codon optimization and gene synthesis
Genscript performed codon optimization for several human TCR alpha and beta chain genes. We sent them the sequence of out original TCR chains (about 2kb total) and they did the rest. After analyzing our sequenced they optimized it for optimal expression and synthesized the genes and sent it to us in a pUC57 vector.
Optimized sequences were used to generate lentivectors encoding the TCR genes to transduced human CD8 T cells
