Our group studies T cells in the blood of untreated chronic HIV+ individuals. We are constantly expanding and optimizing our T cell panels to measure various T cell subsets, activation, and inhibitory receptor phenotype. We pursued using Tim-3-PE in a panel to study inhibitory receptors on CD4 T cells, but ultimately moved to Tim-3-PEDazzle due to the fact that a marker we were interested in was only available on PE.

Results Summary

We first titrated Tim-3-PE alone to determine the volume that yielded the best stain index. 1uL Tim-3-PE had the highest stain index and worked great to detect Tim-3+ T cells, especially after overnight SEB stimulation (1ug/mL).

Tim-3-PE works great in a multi-color panel. Unfortunately PE is a heavily used channel; if you don't have room on PE, consider Tim-3-PEDazzle as an alternative.
