Protein Kinase D (PKD) is family of Ser/Thr kinases that regulate multiple biological functions including cell migration, proliferation, apoptosis, transcriptional activation, angiogenesis, and Golgi organization and vesicle trafficking. Like many other kinases, PKD functions are mediated via phosphorylation of different substrates. The goal of this project was to demonstrate if rhotekin was a bona fide substrate of PKD and whether this phosphorylation is important for regulating RhoA. In this experiment, rhotekin expression was tested in multiple cell lines to determine an optimal cell line for downstream experiments.

1:500 dilution in blocking agent; 16 hr at 4 deg

Goat-anti Rabbit IgG-HRP; 10000 dilution in blocking agent; 1 hr at room temp.
