Bio-Rad's midiprep kit is a scaled up version of their Quantum Prep miniprep kit. Both vary from the standard prep protocols by having the user add the plasmid DNA binding matrix to the plasmid-containing solution, instead of adding the latter to a spin column. Overall the kit is easy to use, rapid, and delivers a good yield of DNA.

Protocol Overview

The kit is comparable to many midiprep kits on the market, but differs in that one adds the DNA binding matrix to the plasmid containing solution that is left following the high speed centrifugation of the bacterial cell debris. The matrix is then centrifuged, washed 1x, and then loaded into a standard spin column for a second wash, followed by elution.

Tips

The spin times recommended are too short. I lengthened them to 10 mins for pelleting the bacteria, 15 mins @ 8000 G for the pelleting of the bacterial debris. I use a smaller elution volume in order to increase concentration.The degraded RNA or nucleotide pool at the very bottom of the gel (see image) can probably be eliminated by adding some RNAse to Buffer I.

Results Summary

On my first try with the kit, my yield in 500 uL of elution buffer was ~220 ug (for .44 ug/ul).

Additional Notes

While some kits recommend against using TB to grow the bacterial culture, I had no problem using this media with this protocol.
