This kit was employed for astroglial identification and quantification after SNC lesion in SNpc with 6-OHDA.
Immunohistochemistry
Brains slices fixed in 4% PFA
Coronal brains slices were extensively washed with PBS, and endogenous peroxidase was inhibited with 3% H2O2. Slices were blocked with 2% BSA, and incubated with primary antibody overnight at 4˚C in blocking solution. After primary antibody incubation, slices were washed in PBS, and incubated in Tracer solution (kit) for 15 minutes at room temperature. Then, slices were washed in PBS and incubated in linker solution (Kit) 15 minutes at room temperature and washed again in PBS. Then, slices were incubated with Diaminobencidine for 10 seconds.
It is really helpful to make standardization every primary antibody, because you can play with Tracer and Linker incubation times
It works nicely, and the kit is recognizes mouse and rabbit primary antibodies.
