Human endothelial cells were deleted for MAPK using its specific siRNA and tested for its effect on growth factor induced Fra2 expression levels in cells. After gene knockdown, cells were growth arrested for 24 hrs and treated with specific growth factor for indicated time. After treatments, cells were lysed in RIPA buffer, whole cell extracts were made resolved in 10% SDS-PAGE and immunoblotted for Fra2 levels using its specific antibody. Fra2 antibody was obtained from Cell Signaling Technology and selection was based on merit indicated in antibody flyer.
