Human retinal endothelial cells were subjected to gene knockdown using specific siRNA. After 48hr of gene knockdown, cells were growth arrested, treated with growth factor and equal amount of protein were resolved in 10% SDS-PAGE followed by immunoblotting with phospho-specific antibodies. Further, immunoblotted nitrocellulose membrane was reprobed with β-Tubulin Ab for to normalize or to test the effect of siRNA on its target and off-target molecules. Selection of β-Tubulin antibody was based on information provided in antibody flyer.

Results Summary

β-Tubulin antibody has detected only specific band of β-Tubulin protein at 55 kDa. However, there was effect of siRNA on its off target molecules.
