Mouse pups were subjected to low oxygen concentration for different time periods. Pups kept at normal room air were taken as control. At specific time periods eyes were enucleated, retinal extract were made, and equal amounts of proteins were resolved in 10%SDS-PAGE followed by immunoblotting and detection phospho-YAP (Ser127) levels using its specific antibody obtained from Cell Signaling Technology

Experimental Design and Results Summary

Applications

Western Blot

Sample

Mouse whole tissue lysate

Primary Incubation

Dilution (1:1000) in 5% BAS prepared in TBST, overnight incubation at 4 degree Celsius

Blocking Agent

5% skimmed milk prepared in TBST

Secondary Incubation

Dilution (1:4000) in 5% skimmed milk prepared in TBST, 1hr at RT

Tertiary Incubation

NA

Detection

ECL detection in X-Ray film

Results Summary

In the present study, antibody from cell Signaling technology was used for immudetection which showed excellent specificity towards phospho-YAP (Ser127) in mouse tissue. Although, a significant response of treatment on phospho-YAP (Ser127) levels were observed.
