The activities of firefly and Renilla luciferases are measured sequentially from a single sample and the Renilla is used as the inter control to calculate the relativity changes on the interaction between two genes. All the operations were done in accordance with the protocol provided with the kit.

After transfection with Renilla and the plasmid of interest in the target cell with lip 2000 or lip 3000 transfection reagent, 36 hours or 48 hours after transfection, collect the cell with the lysis buffer from the kit, and then follow the luciferase kit protocol to check the luciferase activity.

The expression level is dependent on the cell confluence and transfection efficiency.
