While there are a number of methods to separate/sort out lymphocytes from peripheral blood cells, isolating CD4+ specifically with a high degree of purity may prove difficult. The kit by STEMCELL allows for an easy isolation of CD4+ t-cells for the utilization of different assays (e.g. regulatory T-cell assays/induction). While the kit may be expensive, it may be fruitful in situations where a FACS machine is not available.

Protocol Overview

1) Incubate peripheral blood mononuclear cells with 50uL of enrichment media for 10 minutes2) Incubate the concoction with 100uL of magnetic beads for 5 minutes3) Place in magnet for 5 minutes and pour out the unbound CD4+ lymphocytes

Tips

If a high cellular density is required, resuspend the beads in fresh media after pouring out the selected lymphocytes and re-incubate in the magnet for 5 minutes for a 2nd pour

Results Summary

Isolated pure CD4+ lymphocytes with high viability (>95%)
