Our lab focuses on the molecular imaging of cancer with antibodies and nanoparticles. To show that the nanoparticle-based imaging agent is reaching the tumor, we must stain the tissues to determine the spatial location. In addition, we stain the vasculature to determine the imaging agent was able to effectively extravasate. We chose this product because the company provides great customer service.

Tumor tissue from U87mg mouse model

Primary Incubation

Alexa Fluor® 488 anti-mouse CD31 (5 µg/mL) for 1 h at 25 °C

Blocking Agent

10% donkey serum for 30 min at 20-25 ºC

Secondary Incubation

DAPI-containing mounting medium was applied and the slides were cover-slipped

Tertiary Incubation

There was no tertiary incubation.

Detection

Nikon A1R + Confocal Microscope and imaging analyzed using the NIS-Elements Ar with Deconvolution software package

Results Summary

CD31 and DAPI were visualized in the imaging with DAPI represented as blue and CD31 in green.
