The focus of our lab is molecular imaging of cancer with antibody-based imaging agents. To do this, we must digest antibodies to obtain F(ab')2 or Fab. This enzymatic reaction requires a temperature of 37 degrees Celcius with constant agitation. In addition, radiolabeling of antibodies must be done with the same conditions. Currently, we have two of these systems in our lab. We chose this system because Eppendorf has a great reputation for providing elite products that last a long time.

Protocol Overview

We use the Fab or F(ab')2 kit for antibody digestion (ThermoFisher Scientific). After addition of the enzyme, we incubate the samples for 5 hour at 37 degrees Celsius on the Eppendorf ThermoMixer F1.5 at 550 rpm. For antibody radiolabeling, we add the chelated antibody to a 1.5 mL tube with the radioisotope. After changing the pH (isotope-dependent), we place the tube on the Eppendorf ThermoMixer F1.5 for 1 hour at 37 degrees Celsius and 550 rpm.

Tips

The machine is simple to use and it has buttons at the top with pre-set temperatures.

Results Summary

We have successfully digested many enzymes. To confirm the digestion, we run the protein on a gel. In addition, we have never had any problems with radiolabeling. We have never had any problems with the ThermoMixer changing temperatures or speeds. Our last ThermoMixer lasted 9 years and we expect our two new ThermoMixers to last the next decade.
