We analyzed murine lung homogenate supernatant, detection of KC was easy in infected samples.

Lung homogenate
Mice were infected with M. tuberculosis for 3 weeks. Lungs were collected and homogenized in 5mL PBS-T. 1mL of homogenate was aliquoted and spun down at max speed, and the supernatant was filtered through a 0.2um PVDF filter. Sample was diluted 1:3 in the appropriate buffer as per product guidelines.
May need to try several dilutions to get your sample within the standard curve.
Results were consistent and product is very easy to use.
