Our work involves the characterization of mouse macrophages from the peritoneal cavity. In order to gate most effectively the peritoneal macrophages from other cells, we use CD11b and F4/80 as surface markers.

Mouse peritoneal exudate
1:100 dilution of each FITC anti-mouse F4/80 and eFluor 450 anti-mouse CD11b on ice for 10 minutes
1x PBS + 2%FBS + 2mM EDTA

Gating on forward/side scatter alone does not give a uniform population with respect to CD11b and F4/80 levels. Using this color combination allows for a clear distinction between macrophages and other cells in the peritoneal cavity.
