The general aim in using the QIAamp Mini Kit is to obtain reproducible results in our genomic DNA preparations. Genomic isolation is relatively straightforward and there are many kits available to do so. Our lab genotypes mice weekly and needs a fast and reliable kit to obtain high quality genomic DNA. In doing so we can get our screening performed faster and save money in animal housing costs. The Qiagen brand speaks for itself. While slightly more expensive than other kits, the results obtained, waste reduced and time saved more than justify the extra expenditure.

Protocol Overview

Like many of the Qiagen protocols the QIAamp kit relies on anion exchange spin column to bind, wash and then elute the genomic DNA product. After an overnight incubation to remove protein, the DNA is bound and isolated in under 45 minutes. Waste is minimized and DNA obtained is of high quality.

Tips

Have the heat blocks equlibrated prior to starting the protocol. Use no more than 25mg of tissue. Have RNAse A (-DNAse) at 100mg/ml prepped and ready.

Results Summary

High purity genomic DNA with A260/280>1.8 and a A260/230>2 across all samples analyzed. Downstream PCR applications yielding reproducable results.
