One of the goals of my project is to carry out protein-protein interactions with components that are relevant to the hedgehog signaling. A first step in the process is to test the antibodies to determine the expression in either over-expression or endogenous levels. The goal of this experiment was to test the expression level of PKA catalytic subunit in NIH-3T3 cells.

NIH-3T3 whole cell lysate_50 µg

Primary Incubation

1:500 dilution in blocking buffer and incubation for 16 hr at 4 deg on a rotator

Blocking Agent

5% Milk in 1x TBS-T (0.1%)

Secondary Incubation

1:5000 dilution in blocking buffer and incubation for 1 hr at 25 deg on a rotator

Tertiary Incubation

None

Results Summary

This is a fantastic antibody that gives a specific signal at 40 kDa and no non-specific bands. I had used it a couple of times and never it had never failed in my hands.
