Our lab works with cell lysates to study various proteins related to cancer. Many of these proteins are quickly degraded by phosphatases, and for this reason we use phosphatase inhibitors in our lysis buffer. We chose this specific phosphatase inhibitor cocktail because of its concentrated, high-quality inhibitors. It works in combination with the "Set I" Phosphatase Inhibitor Cocktail also sold by Millipore.

Protocol Overview

Dilute 1:100 in lysis buffer just prior to use. We normally use between 50 and 100 uL of lysis buffer per 10cm plate when lysing cells, depending on the confluency of the plate.

Tips

Issue with manufacturing can cause significant shipping delays.

Results Summary

We were able to block phosphatase activity among cell lysates and obtain an accurate picture of the phosphorylation states of proteins in our cell lysates. This product consistently returns on its promise.

While I have had numerous issues with the customer service at EMD Millipore and have my reservations about the manufacturing process at Calbiochem, this product has been of a consistently high quality and performed well even after repeated freeze-thaws. I highly recommend it.
