Following vaccination, the characteristics of the T cell responses induced are of importance. Differing vaccines, routes of immunisations, or adjuvantation alter the antigen-specific T cell profiles. CD44 is a marker of effector-memory T cells, and should be investigated following vaccination. This antibody can distinguish CD44+ T cells from those not expressing the marker.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Murine PBMCs from blood draw.

Primary Incubation

0.5 ul Antibody per 2x 10^6 cells, diluted in MACS buffer in final volume of 50ul.

Blocking Agent

MACS buffer. Incubated for 30 mins at 4C in dark.

Secondary Incubation

NA

Tertiary Incubation

Results Summary

This antibody distinguishes cells expressing high or low amounts of CD44 on murine T cells, in particular CD8+ T cells. Though not disparate populations, distinctions between low and high expression of CD44 can be made.
