Company:

BioLegend

Product Name:

Recombinant Mouse TNF-α (ELISA Std.)

Catalog Number:

575209

We measured the secretion of TNFa in mouse macrophage cells after LPS stimulation in the presence or absence of the fractions/treatment. We quantified the concentration of TNFa using antibody pair and standard from BioLegend. These reagents work pretty well with high sensitivity in our ELISA.

Experimental Design and Results Summary

Application

ELISA to quantify mouse TNFa

Starting Material

Cell culture supernatant at different dilutions

Protocol Overview

Capture antibody was coated (1.5 ug/ml ) overnight and incubated with standard protein for 1 hr at 37 degrees. Then, plate was blocked with blocking buffer and samples of mouse TNFa standard protein at different concentrations were added and incubated at 37 degree for 1 hr. 0.5ug/ml of secondary antibody was added in each well and incubated for 1hr at 37 degrees. HPR-streptavidin were added and incubated for 1 hr at 37 degrees (1 : 10,000). After wash, TMB was used to develop the signal.

Tips

In case the signal is not strong, warm TMB to RT before adding

Results Summary

Using the protein standard combined with antibody pair, the ELISA generated good signal that is sensitive enough to detect TNFa standard protein as low as 1 pg/ml. We found that a fraction from bacteria secretion suppressed LPS induced TFNa secretion in macrophages.

Additional Notes

This standard protein comes with small vials, it's easy to use.

Summary

The Good

Sensitive and easy to use.

The Bad

None

The Bottom Line

We usually use the standard with antibody pair to perform ELISA to quantify TNFa, and we got repeatable reaults. So, the reagent is reliable.
