We are interested in detecting anti-nuclear antigen autoantibodies (autoimmune predisposition) in aged mouse serum and comparing that with our mutant mouse lines. ELISA is the simplest way to measure protein levels in multiple serum samples. This product was selected because of the low cost.

Protocol Overview

1. Add 1:50 diluted mouse serum into each well. Duplicate for each mouse. Kit suggests 1:100 dilution 2. Add Anti-nuclear antibodies (ANA) standard supplied from Kit: 0-50U-200U-500U-1000U/mL. 3. Follow the exact steps described by kit. 4. Measure OD450 and OD630 reads. 5. Calculate ANA standard curve and serum level

Tips

Take care during multiple washing steps.

Results Summary

Duplicate wells show very similar reads and 50U/mL wells still can distinguish from background buffer.
