We have been using mouse GMCSF for many years for cultivating and harvesting bone marrow macrophages and dendritic cells. This product is very costly from all vendors and we have tried many other vendors whose price is a little less to decrease the cost. However the yield of macrophages and dendritic cells that we get from other vendors’ GMCSF was much low compared to the one we get from BioLegend. You will need to use only 10ng/ml of this in the medium to get a very good yield. It would have been nicer though if the cost of BioLegend’s GMCSF were less than others. Certainly this one is the best Mouse GMCSF available in the market.

Protocol Overview

We used 10ng/ml of this product in the IMDM medium and seeded this medium with bone marrow flush of mouse. We then spiked more medium with 2ng/ml every alternate day until day 7 and then harvested all the cells through centrifugation and separated Macrophages and DCs through magnetic bead columns.

Tips

Spiking of mouse bone marrow derived cells with mouse GMCSF every alternate day is very important to get good yield.

Results Summary

Fractionated cells can remain viable in GMCSF for a long period of time (More than 8 weeks) if GMCSF is replaced every week.
