It is often very difficult to distinguish between human M1 and M2 macrophages. This is because there is no universal marker that distinguished one from another. However, certain receptors can be expressed at higher levels in one or the other of these polarized states. CD80 is one such marker. It plays a critical role in regulation of T cell activation, and is hence overexpressed in M1 (but not M2) macrophages. This antibody allows for this distinction to be seen, and will undoubtedly be useful in the armor to distinguish the two polarization states.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Human macrophages

Primary Incubation

No dilution required! Add ~5uL to the sample for 30 minutes on ice.

Blocking Agent

Unlabeled IgG solution

Secondary Incubation

NA

Tertiary Incubation

NA

Detection

Flow cytometry on 695/40 BP filter

Results Summary

The results are encouraging, showing strong staining on M1-polarized cells compared to the M0 and M2 macrophages. This on its own may not be enough, but combined with other M1 and M2 markers, it will help establish whether the cell is an M1 or an M2 macrophage (arbitrary as that may be).
