We use this kit to isolate plasmid from 4 ml bacterial cultures. We generally use this kit to isolate good quality plasmid for validation during cloning procedures. This kit provides us with good quality DNA with no RNA contamination.
Cloning
Bacterial culture
We followed the manufacturer protocol. Briefly, 4 ml of bacterial culture is pelleted, 200microliters of resuspension buffer is added to that to resuspend the pellet and 200 microliters of lysis buffer is added to that and incubated for 5min, next 350 microliter of neutralizing buffer is added and spun down. The sup is loaded on to the column for purification.
Do not extend the lysis step for more than 5 min
Good quality plasmid is obtained
Dry the column to remove the residual ethanol to get good yield.
Very easy to use to obtain a good quality plasmid for cloning purposes
