2x Midas Mix with Thermus aquaticus DNA Polymerase 1

Our lab uses PCR for a number of purposes (cloning, mutation confirmation, etc), mostly with E. coli and Staphylococcal DNA. We have found that this 2x Midas mix works great. It is convenient because all reagents are pre-mixed, simply add DNA, primer, and water. We have tried with both pure DNA as well as "colony PCR" (where we directly add bacteria cells to the reaction) and have gotten results similar to any other Taq polymerase.
PCR amplification of DNA
Bacteria DNA (either a prep or bacteria cells), primers, water
Add DNA, primers, and enough water to dilute Midas Mix to 1x. Place in thermocycler and begin PCR program. Since Midas Mix already has loading dye, you can load onto gel without doing anything else.
Depending on primers and amplification target, you may need to adjust primer levels. You may also need to add additional MgCl2, although we have never had to.
