We are assessing consequences of Aryl hydrocarbon Receptor (AhR) activation in dendritic cells. Specifically for these experiments we were assessing Aryl hydrocarbon Receptor activation in bone marrow derived dendritic cells. Precursor cells are isolated with Lympholyte M from whole bone marrow suspensions and grown in the presence of GM-CSF for 5-7 days. We then use flow cytometry and RT-PCR to assess changes in dendritic cells after AhR activation. We use lympholyte M for the centrifugation step in preparation of bone marrow derived dendritic cells.
Density Centrifugation
Murine Bone Marrow
Suspend bone marrow cells at 15 million/ml in 15ml conical. Slowly pipette 5ml of Lympholyte M beneath the cell suspension. Centrifuge at 2000rpm for 15 minutes with break turned off.
Make sure centrifuge is well balanced
This product gives us a consistent, well-defined buffy coat of cells.
