We use the Bioruptor for our Chromatin immuno-precipitation assays. It is essential for us to optimize the size of DNA to fit our requirements before antibody pull-down. This product allows us to do that.

Protocol Overview

Crosslink the samples with formaldehyde. After the crosslinking, this chromatin is chopped into a specific size of around 500bp using Diagenode' biorupter. As we use macrophage cell line we prefer to give 10 cycles (30 sec on and 30 sec off ). This sample is then taken and run on an agarose gel to confirm the size of chopped chromatin. After that, the pull down experiment will be carried out with specific antibodies.

Tips

Make sure the the sonicator tub is at 4 degrees celsius. If the machine is hot then stop for a while and then restart. Never give more than 10 cycles during a single stretch. Make sure the sample is maintained at lower temperature.

Results Summary

30 sec on and 30 sec off is considered one cycle. By giving 10 cycles we get a DNA of around 200-500BP in length.
