Company:

Thermo Scientific Pierce

Product Name:

SuperSignal West Pico Chemiluminescent Substrate

Catalog Number:

34077

I use this chemiluminescent substrate to detect HRP conjugated secondary antibodies in order to detect proteins via Western blot. I used this chemiluminescent substrate because it is an easy two solution system that has consistently worked to detect HRP labeled secondary antibodies when I perform Western blots.

Experimental Design and Results Summary

Application

Western Blot

Starting Material

HRP labeled secondary antibodies

Protocol Overview

After incubated with HRP labeled secondary antibody, mix the two solutions in the kit 1:1 and then add to your membrane. Take pictures either on X-ray film or digitally.

Tips

Mix solutions well before adding to membrane

Results Summary

The Western blot image was taken digitally and shows a single dark band. This suggests the chemiluminescent substrate is being processed by HRP and producing detectable light.
