Our laboratory works on establishing mouse brain connectomes via tracer/viral stereotaxic injection and subsequent microscopic imaging. We use VS120-S5 routinely for flat mount brain sections. VS120-S5 is able to provide the throughput and high quality image for our purpose.

Protocol Overview

The olympus VS120-S5 is used to scan brain sections for 5 color channels (Nissil, Phaseolus vulgaris leukoagglutinin, fluorogold, biotinylated dextran amines and cholera toxin B subunit).

Tips

Hoechst dye will bleed into fluorogold channel. Do not use simultaneously.

Results Summary

The scope is equipped with robotic stage movement and automatic focusing. Images from individual vision fields were tiled to generate whole brain section images at high resolution.
