Many cytokines are elevated in acute dengue infection and the main aim was to determine if severe dengue could occur due to reduction in type I interferons. In order to answer this question, we obtained blood samples from very early in the disease and at a later time point in a large cohort of patients to determine if reduction in type I interferons were associated with severe dengue. We chose this product because pre-coated plates were available and our previous experiences with this manufacturer was quite good.
Detection of IFN alpha levels in human serum
Serum
Human serum was used and the quantitative ELISA was carried out according to the manufacturer's instructions. We did the standard curves and the samples in duplicate. PRISM was used to calculate the unknowns from the standard curve.
As long as you follow the manufacturer's instructions it's quite simple to carry out.
We did not observe any changes in IFN alpha levels in those with severe dengue when compared to those who had non severe dengue. However, a significant negative correlation was observed with IFNa levels and white cell counts.
