Our lab does a lot of chromatin immuno-precipitations, 3C assays, immuno blots and other applications where you need to incubate the samples at various temperatures for extended durations, sometimes for more than 16 hrs !!! The key to the success of the experiment is to have a constant temperature and continuous shaking for extended period of time and that is why we chose Eppendorf Thermomixer R. I have checked it myself using the thermometer and the temperatures on the machine are constant. Its used extensively and is virtually never free in the lab !!

Sonicated chromatin from formaldehyde cross-likned HeLa cells

Protocol Overview

Sonicated chromatin is incubated with antibodies and subsequently with beads and then washed. The beads then need to be incubated at 65 degree overnight in shaking conditions. Thats where Thermomixer comes in.

Tips

Using the interval mix helps. It will mix the beads for set time and wil be then static for set time and repeats this cycle until you want to stop.

Results Summary

After de-crosslinking, the proteins are digested with proteinase K, samples purified by columns or phenol:chloroform. This DNA can then be used to perform qRT-PCR with primers specific for loci and compare the occupancy of different proteins.
