The human MacroH2A antibody from Active Motif does not work for immunofluorescence experiments in dilutions as low as 1:50, while antibodies from other companies do work at concentrations of 1:500. The IF experiment was put through several optimization experiments to detect MacroH2A in a variety of cell types including female hES/hiPS and 293 cells to visualize heterochromatin surrounding the X chromosome. It was also attempted to perform IF and FISH simultaneously, which works with other antibodies.
