Easy enough to train undergrads on, but powerful enough for a senior research scientist, the Roche LightCycler 480 is the gold standard I use to compare all other PCR instruments to for gene quantification and endpoint genotyping. Fantastic sensitivity and reproducibility both with our in-house primers and when testing other commercial sets when paired with SYBR Green I. Extensive room for expansion with multicolor assay options and high throughput capabilities. It is a high initial investment and has some consumables, but for our lab it's worth the price for high quality results.

Protocol Overview

Conventional 2 step RT-PCR with SYBR Green I quantifying an important translocation in cancer diagnosis and prognosis.

Tips

The usual qPCR advice- aliquot primers, equal reaction volumes, spin the plate before running, and validate validate validate

Results Summary

Used extensively in our lab to both validate cell lines and patient samples before using them in other experiments. High sensitivity and specificity, down to the levels of 10 cells/mL each containing one translocation event.
