As a TCR-immunotherapy project, a part of my research includes cloning various inserts (T cell receptors) into lentiviral vectors as well as a lot of general cloning. I used this product mainly to purify PCR reactions and enzyme digestion reactions.

I use this kit regularly to purify enzymatic reactions. I like that the protocol is very simple, using 30 second long spins rather than 1 minute. And extremely simple to use. My undergrad, who had no experience with even pipetting, picked it up in the first try. I also like that you can elute in 15 ul volume, without losing yield.
Elute in 15 ul for the highest concentration. Also, even though they say dilute reactions to 100 ul, it's not necessary
This kit has consistently given me extremely high yields for DNA cleanup. When I used Promega Wizard / Invitrogen PureLink / Qiagen kits, I almost always got ~50-55% recovery of DNA. With the Nucleospin kit, I get 80-95% recovery. This means that I can use less DNA, hence less enzyme for my RE digests. So this kit ends up saving me money at the end and also time because of its very short protocol. On another note, I have also used it for gel extraction of DNA, and I got 65-70% yield. As a molecular biologist, this was the first time i ever got such a high yield. I would highly recommend this kit for high yield, high-quality, headache-free DNA cleanup/gel extraction.
