c-Myc proteins function in cell proliferation, differentiation and neoplastic disease. Amplification of the c-Myc gene has been found in several types of human tumors, including diffuse large B-cell lymphoma (DLBCL). In DLBCL, high expression of the c-Myc protein has been associated with poor prognosis. I am using the c-Myc antibody to measure c-Myc protein expression levels by immunohistochemistry (IHC) in DLBCL formalin fixed paraffin embedded tissue (FFPET) biopsies.

Mouse monoclonal (9E10) antibody to c-Myc (cat # sc-40). The antibody was used at a 1/30 dilution in Ventana® Diluent (cat #760-108). The protocol comprised Ventana cell conditioning 1 (CC1) (cat #950-124) based pre-treatment step for antigen retrieval, 32 minute antibody incubation, amplification kit (parts A&B, Ventana, catalog #760-080), 8 minute hematoxylin (Ventana, cat #760-2021) and 8 minute bluing reagent (Ventana, cat #760-2037in conjunction with the Ventana iView™ DAB Detection Kit (cat #760-091).
No blocking agent is required using this antibody with the Ventana iView™ DAB Detection Kit.
Affinity-purified biotinylated antibodies: goat anti-mouse IgG, goat anti-mouse IgM and goat anti-rabbit IgG (antibodies are included in the Ventana® iView™ DAB Detection Kit). These antibodies are supplied in a ready-to-use pre-diluted automatic dispenser and the cocktail is applied to each slide.
